It’s hard to tell from your screenshot, but can you confirm if you’re trying to apply the Blend mode to the fill or the entire layer? 

If you’re trying to apply the Blend mode Overlay to the fill, there won’t be any initial noticeable change when applying, but if you want to apply the Blend to the entire layer, you’ll want to add it from the drop icon within the Appearance section.
