I would love that if I change the size between variants in interactive components, like adding some more information on a card in variant 2, would push the content down or sideways, when seen in a prototype.

I agree this would be useful. When you choose one variant in design mode, components are stacked with auto layout but if that variant is triggered in prototype mode, it does not stack.
I agree this as well. Looking forward to see this feature implemented in Figma.
